Исследование Met235Thr полиморфизмов гена AGT, ассоциированных с сердечно-сосудистыми заболеваниями (ССЗ), как ранних молекулярно-генетических предикторов профессиональных заболеваний у квалифицированных спортсменов Республики Адыгея (РА).</p></sec><sec><title>Материал и методы</title><p>Материал и методы. Распределение Met235/235Thr полиморфных вариантов AGT гена исследовано SNP — методом (single nucleotide polymorphism) с аллель-специфическими праймерами и электрофоретической детекцией результатов (НПФ “Литех”). С704Т полиморфизмы гена AGT (rs699) с заменами нуклеотида цитозина на тимин в 704 позиции гена и метионина на треонин (Мet&gt;Thr) в 235 аминокислотной последовательности ангиотензиногена типированы в образцах геномной ДНК квалифицированных спортсменов (n=40), доноров (n=120) и больных с ССЗ (n=64) в возрасте 23-65 лет из двух этнических групп адыгов и русских. Экспериментальные данные проанализированы адекватными статистическими методами (SPSSStatistics 17.0).</p></sec><sec><title>Результаты</title><p>Результаты. Частоты 235Thr аллеля и T hr235Thr генотипа гена AGT статистически значимо повышены в группе больных по сравнению с контролем (р=0,05, χ2=5,84; р=0,01, χ2=6,2) и спортсменами (р=0,05, χ2=6,15; р=0,02, χ2=5,04). Высокий риск развития ССЗ у носителей как аллельного 235Thr варианта (OR=4,34) так и T hr235Thr мутантного генотипа (OR=3,89), обусловленный повышением уровня ангиотензиногена в плазме крови, подтверждает его ассоциацию с болезнями сердечного континуума (БСК) у жителей РА. В условиях интенсивных физических нагрузок Thr235Thr генотип у квалифицированных спортсменов является неблагоприятным фактором, приводящим к кардиоваскулярной патологии.</p></sec><sec><title>Заключение</title><p>Заключение. Полученные новые данные о повышении частоты прогностически неблагоприятного Met235/235Thr полиморфизмов гена AGT ССЗ у квалифицированных спортсменов могут быть использованы в качестве ранних молекулярно-генетических предикторов нарушений в работе сердечно-сосудистой системы на индивидуальном уровне, а также для формирования групп риска с последующей корректировкой тренировочного процесса.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>Aim</title><p>Aim. Assessment of Met235Thr polymorphisms of gene AGT, related to cardiovascular diseases (CVD), as early genetic predictors of occupational diseases in professional sportsmen in Adyghea Republic (AR).</p></sec><sec><title>Material and methods</title><p>Material and methods. Dispersion of Met235/235Thr polymorphic variants of AGT gene studied via SNP-method (single nucleotide polymorphism) with allele-specific primers and electrophoretic detection of results (NPF “Litech”). С704Т polymorphisms of gene AGT (rs699) with replacements of cytosine by thymine at 704 position of gene, and methionine by threonine (Met&gt;Thr) at 235 aminoacid consequence of angiotensine, were typed in the specimens of genomic DNA of professional sportsmen (n=40), donors (n=120) and CVD patients (n=64) aged 23-65 y. o., from two ethnicities — Adyges and Russians. Experimental findings were analized with relevant statistical software (SPSS Statistics 17.0).</p></sec><sec><title>Results</title><p>Results. Prevalence of 235Thr allele and Thr235Thr genotype of AGT is significantly higher in the group of patients comparing to healthy controls (р=0,05, χ2=5,84; р=0,01, χ2=6,2) and sportsmen (р=0,05, χ2 =6,15; р=0,02, χ2=5,04). High risk of CVD development in carriers of allele 235Thr variant (OR=4,34) as Thr235Thr mutation genotype (OR=3,89), caused by the increase of agiotensine level in blood plasma, confirms its association with the diseases of cardiovascular continuum (DCC) in AR inhabitants. Under conditions of intensive physical exertion the Thr235Thr genotype of qualified sportsmen is an adverse factor leading to cardiovascular pathology.</p></sec><sec><title>Conclusion</title><p>Conclusion.
